Abstract

Behind the provocative title “In Amplicon Sequence Variants (ASVs), we cannot trust...", I intend to present why, despite their intrinsic qualities, Amplicon Sequence Variant (ASV) methods are not the Holy Grail of DNA metabarcoding analysis tools, as is often claimed. I will return to what Molecular Operational Taxonomical Units (MOTUs) and ASVs are, explaining how the latter are only a special case of the former, and why we cannot assume that they unambiguously detect true sequences among the experimental noise of the DNA metabarcoding technique.
