Is the sefishness of the Bacillus cereus ATTC 10987 strain caused by the BceS1 type III restriction and modification system?

Several attempts have been made to transform the Bacillus cereus ATCC 10987 strain, but with no success. The nucleotide sequence of an 11kb chromosomal BglII fragment from this strain revealed two closely adjacent open reading frames organized in an operon, of which the deduced amino acids showed 61.6% identity to the type III restriction and modification (R/M) subunits described in G-negative bacteria. The genes were not identified in any of 32 other B.cereus/thuringiensis strains, and several of these strains have successfully been transformed. The system was designated BceS1 as this endonuclease required ATP and Mg2+ as cofactors like other type III endonucleases. Northern blot analysis revealed an enhanced transcription level when the culture was grown in the presence of foreign DNA, and a cell free extract (CFE) from this culture restricted pUC19, whereas a CFE from the same strain grown in plain medium did not. CFE methylation treated pUC 19 was not restricted by the CFE . The CFE methylated the plasmid and protected it against further restriction of the CFE. Evidence for the restriction sites in pUC 19 are demonstrated, and the recognition sequence of the BceS1 are under investigation. BceS1 is the first chromosomal type III R/M system characterized in a Gram positive bacterium.
