Structural Biology of Membrane Proteins in Phospholipid Nanodiscs - from Method Development to Biological Applications 

Membrane proteins play an essential role in cellular signal transduction and the transfer of material across the biological membrane barrier. Due to this fact, membrane proteins account for around 60% of current drug targets and are hence of interest to basic as well as applied research. Unfortunately, little is known about the structure and function of this important protein class. Therefore, we set our focus on the investigation of the structure and functional dynamics of membrane (-associated) proteins using NMR spectroscopy as a main method but also other structural, biochemical and computational techniques. In order to characterize these proteins in a native environment we are using phospholipid nanodiscs, a novel detergent-free membrane mimetic, we recently optimized for NMR spectroscopy and electron microscopy applications.
