Forskningsområde

Jeg er interessert i hvordan genene reguleres; - hvordan cellene holder styr på hvilke av de vel 20.000 genene våre som skal være "av" eller "på" i kroppens ulike celler og vev. Hundrevis av forskjellige transkripsjonsfaktorer bidrar til dette. Men pakking av genenes DNA i kromosomene spiller også en avgjørende rolle for genregulering. Genenes DNA er kveilet rundt histonproteiner til nukleosomer, nesten som perler på en snor, og disse foldes i neste omgang til kromatin i kromosomene. Histonene kan merkes av forskjellige biokjemiske modifikasjoner slik at dette reflekterer genenes status: "av" eller "på". Dersom histonmerkene kopieres og opprettholdes gjennom flere cellegenerasjoner, kan det bidra til å gi cellene en slags "hukommelse"; - cellene "husker" hvilke gener som skal være av eller på. Dette kalles epigenetisk genregulering. For å finne ut hvordan histonene bidrar til genregulering, jakter vi på proteiner som gjenkjenner ulike histonmodifikasjoner og derigjennom deltar i genregulering. Vi benytter både biokjemiske, bioinformatiske og molekylærbiologiske metoder.
