Title
"Polygenic hazard scores for precision cancer screening: from population genetics to clinical implementation"
Abstract
More accurate risk prediction is needed for prostate cancer to identify individuals at greatest risk of early-onset and clinically significant disease. Current screening paradigms, such as prostate-specific antigen screening, pose risks due to over-diagnosis and over-treatment of indolent disease. Polygenic hazard scores (PHS) offer an alternative approach to predicting age-at-diagnosis from genotype data and may be an effective tool for pre-screening risk prediction before symptoms emerge. I will present validation of the latest prostate cancer PHS in two Norwegian cohorts, showing it surpasses rare pathogenic variant screening from whole-genome sequencing, and discuss its integration into two prospective screening trials in the US and Norway.
