Title
“Uncovering the dark proteome and characterizing protein synthesis at scale”
Abstract
Ribosome profiling introduced a new paradigm for studying translation, opening the door for transcriptome-wide analysis of protein synthesis. Despite its potential, individual studies are limited in their ability to uncover translational phenomena due to a lack of sequencing depth and a narrow focus on specific hypotheses. We have processed more than a trillion reads derived from nearly all public ribosome profiling datasets, spanning more than 15000 libraries across more than 1000 studies. We developed an integrated analysis and visualization platform, RiboCrypt, that leverages the power of analyzing thousands of datasets in parallel. Through massive data aggregation, we annotated thousands of translated regions (translons), including 5’ and 3’ extensions of annotated CDSs, translated retained introns, and novel Open Reading Frames (ORFs). Combining this approach with data stratification, we reveal novel phenomena only detectable through the aggregation of data.
