Solubility and dissolution enhancement of poorly soluble drugs
Conferral summary
Main research findings
Nanomaterials offer exciting opportunities to improve drug delivery. For instance, by controlling drug uptake and transport one can optimize therapeutic effects while minimizing side-effects. This thesis investigates the potential of a novel material, silicon nanoparticles synthesized by centrifugal chemical vapor deposition (cCVD), for various pharmaceutical applications.
Characterizations revealed the cCVD particles as either solid or porous “snow lanterns” suitable for carrying drugs. Comprehensive testing showed the particles’ suitability as drug carriers in terms of stability, degradability, non-toxicity, versatility for surface modifications and the ability to tailor the transport of a drug payload.
Drugs with low water solubility, often resulting in poor uptake, showed an enhanced dissolution rate when loaded into the porous cCVD particles. In animal studies, particles formulated with an immunosuppressive drug reduced the side-effects of biological therapies.
This thesis also explored hydrogen gas therapy, which has shown promise as an antioxidant for treating diseases like cancer, cardiovascular and nervous system disorders. The cCVD particles were found to generate hydrogen gas when reacting with water, offering a novel practical method for its administration. Drug-loaded particles could also release hydrogen gas, enabling dual therapeutic effects. Overall, cCVD silicon nanoparticles were shown to be promising for use in several pharmaceutical applications.
