Muscle regeneration in normal and pathological state with focus on metabolism, proteases and cell communication
Conferral summary
Main research findings
Skeletal muscle plays a key role in energy metabolism of nutrients like carbohydrates and fat. Dysregulation of energy metabolism is associated with diseases such as type 2 diabetes, obesity and osteoporosis. Both skeletal muscle and bone are important metabolic organs, and their interaction is important through secreted myokines and osteokines, respectively.
In this work, the proteolytic enzyme legumain was investigated, which has previously been shown to be present in human skeletal muscle cells and to inhibit bone forming cells. However, it was not known whether legumain affected energy metabolism. Here, legumain was shown to play an important role in regulating energy metabolism in both human and mice skeletal muscle cells by regulating several genes and proteins. Furthermore, skeletal muscle cells were shown to influence energy metabolism in bone forming cells and vice versa. These results provide valuable insights into the role of legumain in energy metabolism which could be of important for whole-body energy metabolism.
