Sammendrag
The research for this dissertation was carried out at the Department of Chemistry – University of Oslo.
Human mesenchymal stem cells (hMSCs) play an important role during inflammation and tissue repair. They also have a function as a therapeutic alternative to prevent rejection reactions during allogeneic transplantation. To understand the functions of hMSCs, knowledge of growth and specialization processes of the stem cells is needed. Such knowledge is obtained by studying eicosanoids (signalling molecules) which are produced and released by the stem cells.
Sensitive quantification methods are required because of low concentration of eicosanoids present in the samples. This dissertation is about the development of liquid-chromatography – mass spectrometry methods for determination of eicosanoids. Since the cell culture samples contained many interfering compounds, an interference removal procedure was included in the methods. Still, the analyses could be performed in a relatively short time and with a potential of automation.
