  Liver endothelial cells (LECs) play an important homoeostatic role by removing potentially harmful macromolecules from blood. The extremely efficient endocytosis in LECs makes these cells an interesting model for the study of the involvement of phosphoinositides in the different steps of the endocytic process. In the present investigation we have studied the effect of wortmannin, an inhibitor of phosphatidylinositol kinases, on uptake, recycling and intracellular transport of (125)I-labelled ovalbumin, which is taken up in LECs via mannose-receptor-mediated endocytosis. Wortmannin was found to inhibit both uptake and degradation of ovalbumin. Further studies indicated that the reduced uptake via the mannose receptor was due both to a reduction of the number of surface receptors and a reduction in the rate of receptor-ligand internalization. Transport of ligand from endosomes to lysosomes was prevented, leading to increased recycling of internalized ligand. Wortmannin treatment released the Rab5 effector EEA1 from the endosomes and caused reduced size of early endosomes.

  Expression of Fc(gamma)-receptors (Fc(gamma)Rs) in different liver cells is poorly characterised. In the present study, transcription of Fc(gamma)Rs in different rat liver cells was examined by means of Northern blot analysis and reverse transcriptase-polymerase chain reactions. We found that both Kupffer and liver endothelial cells produce mRNA for Fc(gamma)RIIB2, Fc(gamma)RIII and the gamma-chain, whereas the level of Fc(gamma)RIIB1 mRNA is negligible. In contrast, parenchymal cells produce no Fc(gamma)R mRNA. Copyright 2001 Academic Press.

  Endocytosis may originate both in coated pits and in uncoated regions of the plasma membrane. In hepatocytes it has been shown that fluid phase endocytosis (here defined as 'pinocytosis') is unaffected by treatments that arrest coated pit-mediated endocytosis, indicating that pinocytosis is primarily a clathrin-independent process. In this study we have tried to determine possible connections between pinocytosis and clathrin-dependent endocytosis in rat hepatocytes by means of subcellular fractionation, electron microscopy, and by assessing the influence of inhibitors of clathrin-dependent endocytosis on pinocytosis. As marker for clathrin-dependent endocytosis was used asialoorosomucoid (AOM) labelled with [(125)I]tyramine cellobiose ([(125)I]TC). [(125)I]TC-labelled bovine serum albumin ([(125)I]TC-BSA) was found to be a useful marker for pinocytosis. Its uptake in the cells is not saturable, and any remnants of [(125)I]TC-BSA associated with the cell surface could be removed by incubating the cells with 0.3% pronase at 0 degrees C for 60 min. The data obtained by electron microscopy and by subcellular fractionation suggested that early after initiation of uptake (<15 min) [(125)I]TC-BSA and [(125)I]TC-AOM were present in different endocytic vesicles. The two probes probably join prior to their entrance in the lysosomal compartment. The relation between endocytosis via coated pits and pinocytosis was also studied with techniques that induced a selective density shift either in the clathrin-dependent pathway (by AOM-HRP) or in the pinocytic pathway (by allowing uptake of AuBSA). Both treatments indicated that the two probes ([(125)I]TC-AOM and [(125)I]TC-BSA) were early after uptake, at least partly, in separate endocytic compartments. The different distribution of the fluid phase marker and the ligand (internalised via coated pits) was not due to a difference in the rate at which they enter a later compartment, since a lowering of the incubation temperature to 18 degrees C, which should keep the probes in the early endosomes, did not affect their early density distribution. Incubation of cells in a hypertonic medium reduced uptake both of [(125)I]TC-AOM and [(125)I]TC-BSA; the uptake of [(125)I]TC-AOM was, however, reduced much more than that of the fluid phase marker. This finding supports the notion that the two probes enter the cells via different routes.

  Transcription factors of the STAT family have been implicated in regulation of cell proliferation. EGF activates several STAT proteins in liver. We have studied the relationship between STAT activation and the growth-stimulatory effect of EGF in rat hepatocytes, assessing specific DNA-binding activity of STAT proteins in electrophoretic mobility-shift and supershift assays. In freshly isolated hepatocytes, EGF activated Stat1, Stat3, and, particularly, Stat5b. However, the ability of EGF to produce this activation was rapidly attenuated when the cells were cultured, while the activation by IFN-gamma (Stat1) and IL-6 (Stat3) was sustained. Hepatocytes cultured for 24-48 h are highly sensitive to the stimulatory effect of EGF on S phase entry. In these cells EGF did not detectably activate Stat1, Stat3, or Stat5b but markedly stimulated MAP kinase (Erk1/2). Thus, although EGF has the ability to activate several STAT proteins, this did not seem to be part of the mitogenic mechanisms used by the EGF receptor in hepatocytes.

  Earlier studies have suggested that fluid phase endocytosis in rat hepatocytes takes place via a clathrin-independent mechanism [1,2]. This observation suggests that a relatively large amount of plasma membrane outside coated pits may be involved in hepatic endocytosis. Ricin, which binds to galactose residues on glycoproteins and glycolipids, has, in this report, been used as a general marker for the plasma membrane of hepatocytes. The endocytosis of ricin was compared with that of asialoorosomucoid (AOM) which is taken up exclusively via clathrin-coated pits. Hypertonic medium has been shown to inhibit uptake via coated pits more effectively than clathrin-independent uptake [3-5]. It was found, in this study, that the addition of 100 mM sucrose to the incubation medium inhibited the uptake of 125I-tyramine- cellobiose-asialoorosomucoid (125I-TC-AOM) more extensively than that of 125I-tyramine-cellobiose-ricin (125I-TC-ricin), compatible with the notion that the two probes are internalised via different mechanisms. Subcellular fractionation experiments indicated that 125I-TC-ricin entered a denser endocytic organelle than that receiving 125I-TC-AOM. To determine whether the separation of the two probes was due to a different transport kinetics (i.e. that 125I-TC-ricin is transported more rapidly to a later, denser compartment than 125I-TC-AOM) the cells were incubated at 18 degreesC to allow a slower internalisation/transport of the labelled probes. The results obtained showed, again, that the early endosomes containing 125I-TC-ricin were significantly denser than those containing 125I-TC-AOM. We also employed the horseradish peroxidase (HRP)-diaminobenzidine (DAB) density shift technique of Courtoy et al. [6] to determine whether 125I- TC-ricin and 125I-TC-AOM were in separate endosomes early after their uptake. The results showed that early endosomes containing 125I-TC-AOM were density shifted whereas those containing 125I-TC-ricin were unaffected by the density shift procedure. The use of probes labelled with 125I-TC allowed us to identify compartments involved in the degradation of 125I-TC-AOM and 125I-TC-ricin, by measuring acid soluble radioactivities in the gradient fractions. It was found that 125I-TC- ricin was degraded mainly in endosomes, whereas 125I-TC-AOM, as expected, was degraded mainly in lysosomes.

  Abstract. The effect of sequential combined hormone replacement therapy on the susceptibility of low density lipoprotein to oxidative modification was investigated in a double-blind, randomized placebo-controlled study. Hypercholesterolemic, postmenopausal women were supplemented with 17ß-estradiol and norethisterone acetate (n=13 subjects) or placebo capsules (n=15 subjects) for 12 weeks. They were instructed to follow the American Heart Association step one diet. Low density lipoprotein, isolated before and after treatment was subjected to copper-catalysed lipid peroxidation. There were no significant differences between low density lipoprotein from the hormone replacement therapy and placebo groups, as assessed by measuring the lag time for formation of conjugated dienes, the rate of formation and the amount of conjugated dienes formed, the amount of lipid peroxides generated, and the relative electrophoretic mobility at baseline and after treatment. Dietary records showed that the subjects were consuming similar amounts of fat and vitamins. No major differences were found in the fatty acid pattern of low density lipoprotein from the two groups. In conclusion, the results indicated that hormone replacement therapy with 17ß-estradiol sequentially combined with norethisterone acetate in non-smoking, hypercholesterolemic, postmenopausal women had no protective effect on the susceptibility of low density lipoprotein to copper-catalysed modification in vitro.

  Endocytosis is a mechanism for uptake of material into cells by invagination of the plasma membrane and formation of intracellular endocytic vesicles. The endocytic process may be divided into pinocytosis and phagocytosis. Pinocytosis (“cell drinking”) is a type of endocytosis in which soluble materials are internalized, whereas phagocytosis (“cell eating”) is endocytosis of particulate material. In the present book the term endocytosis is equivalent to “pinocytosis”, whereas phagocytosis means uptake of particles such as bacteria. The endocytic pathway consists of several transport routes: Endocytosed material may be recycled back to the cell surface, it may be transported across the cells, or it may be brought to the lysosomes for degradation. The hepatocytes are polarized cells with a basolateral surface facing the Space of Disse and an apical membrane surrounding the bile canaliculi. Transport of selected material takes place from basolateral to apical side of the cells. Transcytosis in hepatocytes will be discussed in chapter 1. A physiologically important type of recycling is mediated by the neonatal Fc receptor. It binds endocytosed IgG and serum albumin at low pH in endosomes and brings these ligands back to the cell surface. This process, which explains the long half life of these proteins, is examined and explained in chapter 2. The molecular mechanisms whereby transport via the endocytic pathway from the basolateral membrane to the lysosome of hepatocytes takes place is described and discussed (chapter 3), and Prof. Seglen, who discovered and coined the name amphisome, examines and discusses the relation between the endocytic and the autophagic pathways (chapter 4). The liver plays a key role in lipoprotein metabolism. In addition to synthesizing lipoproteins it also express receptors that mediate endocytosis of lipoproteins such as low density lipoproteins and chylomicron remnants. The role of the liver in regulating turnover of lipoproteins is examined and discussed (chapter 5). The liver is the only organ that can rid the blood of cholesterol. High density lipoprotein (HDL) transfers cholesterol from peripheral tissues to the liver (reverse cholesterol transport). Hepatic endocytosis may be involved in the release of cholesterol esters from HDL into hepatocytes. Reverse cholesterol transport is the topic of chapter 6. Hepatic endocytosis plays important roles both in innate and adaptive immunity. Liver cells cooperate to remove unwanted and potentially harmful material from blood. The material may be modified self molecules, foreign molecules or cells entering portal blood from the intestines. Hepatic endocytosis of desialylated glycoproteins were among the very first studies of receptor-mediated endocytosis. Dr. Tolleshaug who did pioneering work in this field reviews work done on receptor-mediated endocytosis of asialoglycoproteins (chapter 7). The liver holds the largest resident macrophage population in the body, the Kupffer cells, which are professional phagocytes. Phagocytosis in Kupffer cells, and the consequent inflammatory and anti-inflammatory processes, are described and discussed (chapter 8). The sinusoidal endothelial cells are very active endocytic cells and express receptors that mediate endocytosis of a large number of modified endogenous molecules and exogenous harmful material. Receptor-mediated endocytosis of the endogenous ligand, hyaluronan, has been studied in great detail, and this work, and endocytosis of other biomolecules, is discussed and reviewed (chapter 9). In chapter 10 the editors try to give an overview of the endocytic function of liver sinusoidal endothelial cells. Kupffer cells, stellate cells, and sinusoidal endothelial cells express both MHC class I and II and are able to prime T cells. Moreover they contribute to hepatic immune tolerance, an issue that is discussed in chapter 11.
