Faglige interesser

Forskningen i min gruppe går i hovedsak ut på å karakterisere vår musemodell som mangler uttrykk av NCU-G1 proteinet, et protein med ukjent funksjon. Formålet er dels å forstå mekanismene som fører til musens fenotype, leverfibrose, og dels via disse studiene å klarlegge NCU-G1s fysiologiske funksjon.

Leverfibrose er et stort og økende helseproblem. Denne tilstanden inntreffer ved varig skade på leveren og kan utvikle seg til cirrhose og kreft. Fjerning av skaden kan føre til reversering av fibrosen, men i de fleste tilfelle er dette ikke mulig. Det finnes i dag ingen medisiner som tilbyr god behandling og dette er derfor er svært aktivt forskningsfelt. Potensielle medisiner må imidlertid testes før kliniske undersøkelser og her brukes dyremodeller. De eksisterende dyremodellene er imidlertid ikke optimale. NCU-G1 ”knockout” musen anses som en potensiell ny forbedret dyremodell for fibrosestudier.

NCU-G1 ”knockout” musen ble skapt ved innsetting av en ”gene-trap” i første intron som fører til stopp i transkripsjonen etter første ekson. Musene har ingen uttrykk av mRNA eller protein for NCU-G1. De er friske, vokser og formerer seg normalt og har som eneste fenotype leverfibrose. Analyserer av lever viser kraftig fibrose med aktiverte Kupffer og stellatceller samt døende hepatocytter. Det arbeides med å finne årsaken til fibrosen i disse musene og dermed antakelig også funksjonen til NCU-G1.

Bakgrunn

Jeg er utdannet ved Universitetet i Oslo og Universitetet i København. Min phd-grad var et studium av endocytose i non-parenkymale leverceller fra rotte. Etter phd-graden arbeidet jeg som forsker ved Institutt for medisinsk biokjemi, UiO, inntil jeg i 1995 tiltrådte stillingen som professor ved IMBV. I løpet av disse årene har jeg hatt flere forskningsopphold av variende lengde ved Laval Universitet, Québec, Canada og et lengre opphold ved Vanderbilt Universitet, Nashville, USA. Min gruppe er medlem av MURES (Muskelforskningsgruppen), et satsningsområde ved Matematisk-Naturvitenskapelig fakultet.
